Fight as Sara, Bun Bun, Jacky, and Jeffry:

To fight as Sara, Bun Bun, Jacky and Jeff, complete the game with each character on normal or greater difficulty.

Fight as the boss

To fight as the boss, set the difficulty level to hard, number of rounds to 2 or more, then complete the game in single-player mode.

big-headed fighters

For big-headed fighters, enable all characters (including the boss and four bonus fighters), then use each of these characters to complete the game. The Big Head Mode option will then be available.
