To fight as Mr. Big, reach him in the single-player story mode, then intentionally lose the match. Restart the game in two-player mode and Mr. Big will now be unlocked at the character selection screen.

Fight as Mr. Karate

To fight as Mr. Karate (the "?" opponent), reach him in single-player story mode, then intentionally lose the match. Restart the game in two-player mode and Mr. Karate will be unlocked, replacing the "?" previously displayed on the character selection screen.
